Slow controlled and delayed release preparations

Controlled release preparation: refers to a preparation in which a drug can be automatically released at a predetermined speed within a predetermined time, maintaining a constant blood drug concentration within an effective concentration range for a long time. Broadly speaking, controlled-release formulations include drugs that control their speed, direction, and duration, as well as targeted and transdermal absorption formulations.

③ Thickener: prolongs the efficacy of liquid medicine. Gelatin PVP、CMC、PVA、 Dextrose. The method of achieving slow and controlled release using diffusion principle:

① Coating: small pills or tablets coated, uncoated, or coated with different thicknesses of clothing.

Blockers: hydrophobic polymer substances, paraffin wax, * * * fatty acids, alcohols, monoglycerides of stearic acid, stearic acid, magnesium, calcium salts, cordyceps.

② Make microcapsules: The microcapsule membrane is a semi permeable membrane, forming a saturated solution for diffusion. The determining factors of drug release rate include the thickness of the capsule, the pore size of the micropores, and the curvature of the micropores.

③ Making insoluble skeleton sheets: using insoluble plastics such as non-toxic polyvinyl chloride, polyethylene, polyethylene acetate, poly (methyl methacrylate), and silicone rubber as the skeleton, suitable for water-soluble drugs. The determining factors of drug release rate include drug solubility, skeleton porosity, pore size, and pore curvature.

Preparation method: Mixing drugs with plastics; Wet granulation; Solvent based granulation and compression.

④ Increase viscosity to reduce diffusion rate: mainly used in injectable and other liquid formulations. Gelatin is used for heparin, CMC (1%) is used for procaine hydrochloride injection (3%).

⑤ Make plant emulsion: Melt insoluble drugs and pour them into the model to form, usually without adding excipients.

⑥ Made into medicinal resin: cation exchange resin is exchanged with organic amines, anion exchange resin is exchanged with organic carboxylates or sulfonates to form medicinal resin. Make capsules or tablets. Only dissociative drugs are suitable for making pharmaceutical resins. Due to limited exchange capacity, high-dose drugs are difficult to produce into drug resins.

⑦ Make emulsions: Water soluble drugs can be made into W/O emulsions. Water phase → oil phase → body fluid.
